The 38-year-old actress, who starred as Vanessa Gold in EastEnders, will play the mother of three tearaway children in the award-winning school drama, the Daily Mirror reports.
Her character, Carol Barry, has to cope with the kids alone because her husband is behind bars.
Mum-of-one Zoe, who is currently shooting her new role, said the family would "ruffle plenty of feathers in Waterloo Road", adding that "you can't trust them as far as you can throw them!"
The former Footballers' Wives star continued: "They're a family of great characters - but you just wouldn't want the Barrys in your neighbourhood!"
Zoe joins another former EastEnder in Waterloo Road as Laurie Brett, who played Jane Beale, has also joined the series. She will star as English teacher Christine Mulgrew in the next series of the BBC drama.
